Universal Avian Reovirus PCR

Type of assay: Real-time polymerase chain reaction (PCR).

Purpose of the assay: To detect Reovirus in turkey and chicken samples

Species: Turkey and chicken

Specimens: Tendon, feces, egg yolk, and cell culture. Other sample types can be tested but have not been validated in our laboratory and will be reported with a disclaimer.

Turn Around Time: PCR will be reported within 1 business day of testing which will take place on Fridays

Analytical sensitivity (1:10 dilution series, 2 dilutions beyond endpoint, tested in triplicate):

  • Reportable range: Positive Ct ≤ 37;  Suspect Ct 37.01 – 39.99;  Negative Ct ≥ 40
  • Limit of detection: 6.2 viral copies/µL

Analytical Specificity: No cross reactivity observed when the PCR was tested against 10 viral isolates (Metapneumovirus, Quail Adenovirus, Adeno 3, Infectious Laryngotracheitis (ILT), Chicken ILT, Turkey Coronavirus, Infectious Bronchitis Virus, Avian Influenza A Virus, AIV H9N9 NIOSH, and Newcastle Disease Virus)


  • Intra-assay: 3.0% coefficient of variation (CV; n=5)
  • Inter-assay: 2.8% CV (n=7)

PCR Assay performance:

Diagnostic Sensitivity (DSe) and Diagnostic Specificity (DSp) (comparison to gold-standard viral isolation; n=176)

  • DSp = 100%  (DSp is the ability to detect true negative samples)
  • DSe = 100%  (DSe is the ability to detect true positive samples)

Note: This assay is expected to detect Reovirus in pigeon and quail samples but has not been validated for these species. The assay has been tested and expected to detect Reovirus in spleen, liver, heart, intestine, tissue homogenate, pancreas, synovial fluid, joint swab, egg shell, and BHI media but has not been validated in these sample types. Unvalidated species and sample types will be reported with a disclaimer.
